Description
Pioglitazone Hydrochloride is the hydrochloride salt preparation of Pioglitazone, a specific ligand of PPAR-γ (peroxisome proliferator-activated receptor-γ). Pioglitaze is a thiazolidinedione compound described to produce antiinflammatory and antiarteriosclerotic effects.
Pioglitazone is demonstrated to prevent L-NAME-induced coronary inflammation and arteriosclerosis and to suppress increased TNF-α mRNA produced by aspirin-induced gastric mucosal injury. Pioglitazone Hydrochloride is an activator of PPAR γ.
Pioglitazone is a prescription drug of the thiazolidinedione (TZD) class with hypoglycemic (antihyperglycemic, antidiabetic) action to treat diabetes. While pioglitazone does decrease blood sugar levels, studies on the main cardiovascular outcomes have not yielded statistically significant results.
Its cardiovascular safety profile compares favorably with that of rosiglitazone, which was withdrawn from some markets after concerns about an increased risk of cardiac events. Pioglitazone has been found to be associated with bladder tumors and has been withdrawn in some countries.
Medical Uses
Pioglitazone is used to lower blood glucose levels in the treatment of diabetes mellitus type 2 (T2DM) either alone or in combination with a sulfonylurea, metformin, or insulin.The main study that looked at the medication, however, found no statistically significant difference in the main cardiovascular outcomes that were looked at.
The secondary outcome of death from all causes, myocardial infarction, and stroke were lower. Pioglitazone has also been used to treat non-alcoholic steatohepatitis (fatty liver), but this use is presently considered experimental.
How should I use this medication?
The usual starting dose for adults is 15 mg or 30 mg taken once daily, with or without food, at the same time each day. Diet and exercise programs should be continued while taking pioglitazone. Your doctor may decide to increase the dose of pioglitazone to 45 mg daily if lower doses do not sufficiently reduce your blood sugar levels. The use and dose of pioglitazone for children must be determined by their doctor.
Pioglitazone may be taken with or without food, and should be taken daily and at regular intervals as prescribed by the doctor so that blood levels of the medication always remain in an effective range.
